Abstract
Cancer is a serious disease with a complex pathogenesis, which threats human life greatly. In our previous studies C3-Pyrazole/Chalcone Linked Beta-Carboline Hybrids were synthesized and evaluated for their cytotoxic activity in a panel of five human cancer cell lines and the new molecules like 7d and 8d were considered as potential leads. These observations prompted us to investigate the apoptotic mechanism underplaying the cytotoxic activity of these compounds in human breast cancer cells MCF-7. In the present investigation studies like flowcytometry of mitochondrial membrane potential (ΔΨm), generation of reactive oxygen species (ROS) and Annexin V-FITC assay revealed that these compounds induced mitochondrial mediated apoptosis in human breast cancer cells (MCF-7). It was further confirmed by western blot analysis of pro apoptotic protein Bax and anti apoptotic protein Bcl-2.
