Abstract
Research about isolation, characterization, and identification of DNA’s lactic acid bacteria of dadih Lintau Tanah Datar District which is antibiotic resistant has been done. Eleven isolates were isolated; U4, U5, U6, U7, U8, U9, U10, U11, U12, U13 and U14; characteristic of isolates are broken white, round shape, convex, edge slick and Gram positive bacteria. Nine (9) isolates are rod cell, 2 others are cocci. isolates were characterized based on antibiotic resistance. Ampicillin 10 μg (AM 10), Diclixacillin 1 μg (DCX1), Gentamicin 10 μg (CN 10), Cofeperazone 75μg (CFP 75), Carbenicillin 100μg (CB 100) have been used as antibiotics test. The result showed that six isolates resistance to all of antibiotics; U4, U5, U6, U7, U8 and U11. The result of DNA identification by primer 27F-1429R, showed that PCR product about 1500 base pair. BLAST analisys result from all dadih Lintau isolate include the lactic acid bacteria group, match 97% - 100% with Lactobacillus plantarum.
